靶向载体通过在编码小鼠NtsR2基因3'端末端序列的终止密码子和polyadenylation位点间插入一个IRES-Cre，同时在IRES-Cre上游放置一个Frt-flanked NEO cassette构建。通过同源重组正确重组的ES克隆被扩增后，注入到小鼠囊胚中以产生嵌合体雄性。Frt-flanked NEO cassette的存在会阻止Cre表达，直到遇到FlpO。(来源：J:263745)
