A mature B cell, where the the immunoglobulin kappa chain complex had undergone Igk-V to Igkj5 recombination, was isolated and reprogrammed to an iPSC (induced pluripotent stem cell). This iPSC was then used to create a homozygous mouse line where all cells contain this allele. The mouse from which the B cells were isolated is a reprogrammable mouse containing the Col1a1tm1(tetO-Pou5f1,-Klf4,-Sox2,-Myc)Hoch and Gt(ROSA)26Sortm1(rtTA*M2)Jae alleles which allow for the reprogramming to be induced by doxycycline.
