PCR primers with flanking restriction sites were designed to amplify the rearranged Valpha3.1Jalpha40 and Vbeta2.1Jbeta2.1 segments from genomic DNA of the Ob1A12 T cell clone. These DNA segments were then subcloned into murine TCR-specific regulatory elements containing genomic TCR expression cassettes pTalphaCass and pTbetaCass, respectively, and microinjected into fertilized oocytes. Mice with this transgene express human anti-MBP (myelin basic protein) 85-99/DR15 T cell receptors. (J:90756)
