Exon 2 of the gene was targeted for flanking insertions of loxP sites. lssDNA (long single-stranded DNA) containing sequence for a loxP site, the end of intron 1, the entire exon 2, the start of intron 2 and a second loxP site was microinjected into zygotes together with two sgRNAs and Cas9 mRNA (with 81 bp poly(A) tail appended). This conditional-ready allele was found in some of the F1 offspring of F0 founders 16 and 17. (J:261995)
