Exon 4 of the gene was targeted for flanking insertions of loxP sites. lssDNA (long single-stranded DNA) containing sequence for a loxP site, the end of intron 3, the entire exon 4, the start of intron 4 and a second loxP site was electroporated or microinjected into zygotes together with two sgRNAs and Cas9 mRNA (with 81 bp poly(A) tail appended). This conditional-ready allele was found in some of the F1 offspring of F0 founders 3, 20, 22 and 23. (J:261995)
