BAC RP23-20F21被用来依次定位Hoxa9和Hoxa10。在每个基因中，插入了一个由lox66和lox71单次只出现的kan/neo cassette，位于第1外显子。这个插入引入了小的缺失/框架移位。在Hoxa9中，通过Cre介导的重组去除了选择 cassette，然后才针对下一个基因。因此，最终靶向载体中只有Hoxa10保留了选择 cassette，之后通过Cre介导的重组将其去除。（来源：J:262635）
