一个反向倒置的条件性内含子(COIN)，包含lox66_HA-Egfp- polyA_lox71序列，通过同源重组在第34外显子编码赖氨酸2384的AAG密码子后以反义方式插入。第34外显子被分隔为上游的PEST区域和下游的Notch2转录激活所需域之后。两个lox位点呈互补方向，因此通过Cre介导的重组会不可逆地反转插入序列。此外，FRT引导的UBp-neomycin选择性载体插入在COIN内含子之后。在Cre重组之前，通过前体mRNA的剪接去除COIN模块，产生与野生型mRNA无区别的转录本。通过Flp介导的重组移除了neomycin选择性载体。(来源：J:246017)
