在exon 4的上游插入了loxP位点，通过同源重组，又在exon 4的下游插入了FRT标记的neo cassette和另一个loxP位点。通过Flp介导的重组，neo cassette被去除了。随后，Cre介导的重组在生殖细胞中删除了exon 4，导致了框架突变。(来源：J:235649)
