在 intron 1 中插入了 loxP 突变位点，neomycin 抗性基因座被 FRT 端口包裹，随后在 intron 3 内也插入了另一个 loxP 点。通过后续的 Flp 介导重组，删除了 neo 骨架。接着，通过 Cre 介导的重组，删除了 exons 2 和 3。exon 2 包含起始的 ATG 译码位点。海马粗制的突触膜片段的免疫印迹确认了这个等位基因没有蛋白质表达。（来源：J:255015）
