在exon 1上游插入了loxP位点，接着是含诺卡因抗性基因座的FRT序列，再以另一个loxP位点为结尾，位于exon 2之后。exon 2下游还插入了经过点突变改造的内源基因cDNA，将Asp34的密码子替换为Asn（p.D34N或p.Asp34Asn）。通过后续的flp介导重组，删除了诺卡因抗性基因座。这就产生了一个条件性可操作的等位基因，它包含1到2号外显子（exons 1-2）都被敲除。只有在内源基因经cre介导删除后，编码为无活性蛋白版本的突变cDNA才会被表达。（来源：J:259970）
