在Y470A基因突变插入到了蛋白产物的去乙酰化酶活化域内的第13号外显子后，还插入了一个Neomycin筛选 cassette。这个筛选 cassette位于目标外显子下游。通过从正确靶向的ES细胞克隆出的始祖小鼠与EIIa-cre转基因小鼠杂交，我们移除了Neomycin筛选 cassette。预测这个突变会干扰Ncor2蛋白与Hdac3去乙酰化酶之间的相互作用。通过免疫印迹分析，对纯合肝脏样本的分析显示，突变等位基因的表达与内源性表达相似。实验表明，突变的Ncor2蛋白不能与Hdac3蛋白发生免疫沉淀实验中的相互作用。(来源：J:252085)
