在ES细胞中，通过同源重组插入了带有tdTomato/T2A/Cre-ERT2/bGHpolyA的磁带，其起始密码子与小鼠Mex3a的exon1对应。筛选出阳性克隆后，这些克隆经FlpO质粒转染以去除neo cassette。阳性克隆被选中，然后进行囊胚注射以产生嵌合体。成功传代至生殖系统的嵌合体与C57BL6小鼠杂交，产生后代用于最终的鼠群繁育。(来源：J:253826)
