从BACPAC资源中心的KOMP项目库中克隆了HTGRS6006_A_G10，这个克隆用于靶向胚胎干细胞。 cassette包含FRT序列、lacZ序列、一个loxP位点，以及一个由人类β-actin启动子驱动的neomycin，后面跟着SV40polyA，再是第二个FRT位点和第二个loxP位点。第三个loxP位点插入在目标exon3下游。exon3被loxP位点夹在两侧。通过在携带这种等位基因的鼠中表达flp重组酶，创建了一个条件性准备（floxed）等位基因。随后的cre表达会导致敲除鼠，移除exon3。exon3的剔除会导致第80个密码子后的帧移。 Western blot分析证实了睾丸中没有检测到全长蛋白。 (J:238496)
