从BACPAC资源中心克隆HTGRS6006_A_G10，使用了KOMP项目 Knockout Mouse Repository中的靶向胚胎干细胞。这个载体包含一个FRT序列，后面跟着lacZ序列和一个loxP位点。第一个loxP位点后面跟着由人类β-actin启动子驱动的neomycin，以及SV40polyA，然后是一个第二个FRT位点和第二个loxP位点。第三个loxP位点插入在目标exon3下游。因此，exon3被loxP位点夹在两侧。通过在携带这种等位基因的鼠中表达flp重组酶，创建了一个条件性准备（含flipped）的等位基因。(来源：J:238496)
