这个靶向载体是通过重组技术构建的。从BAC克隆RP23-RP23-365K8中获取了8528bp的mouse Penk基因最后一个外显子的基因组DNA片段，然后插入到含有DT基因的载体中。在终止密码子TGA和3' UTR之间插入了IRES-Cre frt-PGKNeo-frt的 cassette。通过与表达野生型FLP1的纯合雌性转基因小鼠杂交，删除了neo cassette。（来源：J:253245）
