在小鼠基的细菌人工染色体(BAC bQ73b10)中，插入了编码mCherry和改良型cre重组酶(crei)的序列，两者由病毒T2A序列分隔，以实现双顺式表达。起始密码子在exon 1的外显子和终止密码子在exon 2的内含子之间替换了原有的基因序列。最初产生了4个杂交体(SST-Cre33, -38, -43和-44)，其中SST-Cre-43无法将转录本传递下去。随后，SST-Cre-33和-38这两株失去了这种积极的报告基因表达。最终，仅存的SST-Cre-44被回交到C57B6背景中，历经了8代。(引用文献: J:239793)
