L1L2_gt0 cassette被插入到关键外显子(第7号外显子)的上游，插入位置在chrX:42,222,777[GRCh38构建]。这个L1L2_gt0 cassette包含FRT位点、小鼠En2内含子/splice接受器/外显子(En2 SA)、自切割的2A肽、β-半乳糖苷酶基因(lacZ)、第二个自切割的2A肽、诺如霉素抗性基因(neo)、SV40poly(A)信号，第二个FRT位点以及一个loxP位点。在第7号外显子下游，chrX:42,223,727位置再次插入了另一个loxP位点，这样就将7号外显子两侧都包裹了loxP位点。通过Flp介导的重组，去除了带有FRT标记的neo cassette，使得7号外显子被-loxP标记。(参考文献J:173534)
