在ES细胞中，通过同源重组技术，将一个icre/ERT2 cassette紧接着一个Frt-flanked neo selection cassette插入到了起始密码子ATG之后的顺位。经过定向筛选的正确克隆ES细胞被注入C57BL/6的囊胚细胞。随后，这些小鼠与表达Flp的实验性小鼠杂交，以删除neo cassette。(来源：J:201085)
