使用了BAC(细菌人工染色体)文库，获得了一个包含完整人类糖原醛-3-磷酸脱氢酶(GAPDH)基因的110kb BAC。在这个BAC中，插入了loxP介导的停止信号，后面跟着人类 Catalase(CAT)基因，以替换GAPDH的3-7个外显子。CAT的起始甲硫氨酸被删除，并在氨基端添加了 ornithine转氨酶(Otc)的引导序列的前25个氨基酸，以将这个蛋白质定向到线粒体。(来源：J:183593)
