A loxP site flanked neomycin resistance gene cassette was inserted upstream of exon 1. A 15 amino-acid Avi tag was inserted in-frame with the gene so that the resulting translated peptide would have an N-terminal biotinylation tag. The insertion of the neo cassette is intended to interfere with transcription of the gene and was shown by RT-qPCR to indeed reduce expression to around 27% of wild-type. (J:240473)
