在exon 2上游插入了一个loxP位点，接着在exon 3内61个bp处又插入了另一个loxP位点。通过在intron 1内插入了由FRT引导的抗氨苄青霉素抗性基因座，实现了基因的替换。通过Flp介导的重组，移除了neo基因座。序列分析显示，exon 2至大部分exon 3区域通过Cre介导的重组被删除。 Western blot实验确认了在野生型小鼠脑、肺、肝、胰腺和胸腺的提取物中，没有蛋白质表达。(来源：J:238436)
