Nonchemically modified deoxyribonucleotides were used to introduce the (5-CAAGGCGATCCAGAGAATCAGGACCGGGAAAAGGTAAAGTC-3) mutation into a mouse ES cell line. This alteration is an insertion of 4 nucleotides (AGGA) to generate the Brca15382stop mutation, which resulted in premature protein truncation at aa 1713 and closely resembled the human BRCA15382insC mutation.
