A codon-optimized IRES-MercreMer (MercreMer: cre recombinase fused to two modified estrogen receptor ligand binding domains at both ends) fused gene and a frt-flanked neomycin selection cassette were inserted into the 3' untranslated region (20 bp after the stop codon) of the Kit gene in embryonic stem cells by homologous recomibination.
