靶向载体通过定点突变设计，插入一个T(CG)到A(CG)的替换点突变，随后在第1外显子下游290bp位置插入一个由FRT引导的诺卡因 cassette。这个突变导致第32位的丝氨酸（Ser32）被Thr（苏氨酸）替代。通过Flp介导的重组，FRT引导的诺卡因 cassette被移除。该突变不影响磷脂酶A2的活性，但会影响PRDX6蛋白在肺上皮细胞内膜囊泡和溶酶体的运输（来源：J:237406）。
