The construct is designed to contain (from 5'to 3') an attB site, a CMV-IE enhancer/chicken beta-actin/rabbit beta-globin hybrid promoter (CAG), a loxP-flanked stop cassette (LSL), a myelocytomatosis oncogene (Myc) cDNA with a T58A mutation fused to an internal ribosome entry site (IRES) followed by a luciferase gene, a woodchuck hepatitis virus post-transcriptional regulatory element (WPRE; to enhance the mRNA transcript stability), and a polyadenylation sequence. This construct was microinjected, along with the PhiC31 integrase mRNA, into the pronuclei of zygotes collected from the TARGATT mice on a FVB/N congenic background. TARGATT mice carry the H11 allele ( Igs2tm3.1Luo) or H11P3; a unique sequence from the promoter of the yeast his3 gene followed by three shortened tandem attP sites and one FRT5 site. One founder mouse carried the correctly targeted transgenic insertion of the construct into the first and second attP sites of the locus. (J:101977)
