针对基因的Exons 3和4被替换为一个构建，内容包括：loxP-FRT-polyA-neomycin（反向转录方向）-FRT-Exons 3-9的Pofut2迷你cDNA-polyA-loxP-Stop-mCherry-polyA。通过Flp介导的重组，FRT标记的neo cassette被去除了。这个等位基因不产生mCherry荧光。通过Cre介导的重组，loxP标记的Exons 3-9也被移除。体内原位杂交和HE染色结果显示，胚胎无法表达胎外中胚层和内胚层分化相关的标记。（来源：J:234992）
