一个包含Sox10基因座226kb区段的鼠细菌人工染色体克隆RP24-85014通过重排技术被修改，以表达携带PCWH综合症致病3'端延伸突变(S1)的人类SOX10蛋白，同时插入了荧光标记的Venus蛋白，使其在Sox10起始密码子处融合。在Sox10cDNA下游插入了一个FRT引导的neo cassette。这个突变删除了从可能的终止密码子第二個碱基开始的12个碱基，消除了终止密码子，使得翻译延伸至3' UTR。这为野生型蛋白序列增加了82个氨基酸。生成了两条转基因品系，55号和196号。55号品系携有两个非同源的转基因拷贝，其中一个携带单拷贝，另一个含有两个串联拷贝。（来源：J:227442）
