使用含有FRT位点周边NEO筛选 cassette的靶向载体，插入了位于目标基因exon 2上游和下游的loxP位点。这个筛选 cassette被置于目标基因exon 2之后，又在exon 2上游插入了另一个loxP位点。通过Flp介导的重组去除了筛选 cassette，随后Cre介导的重组剔除了exon 2。原位杂交证实了大脑中没有完整mRNA。(来源：J:232709)
