将含Sox10基因座的BAC（Bacterial Artificial Chromosome，BAC）RP23-28011改造，构建了转基因。构建载体包含改良的cre重组酶（icre）融合的突变雌激素受体（ERT2）编码cDNA序列、SV40polyA序列和Frt标记的PGK-neo cassette，通过细菌细胞的同源重组，这些序列替换Sox10基因的第3外显子。通过诱导阿拉伯糖表达的Flpe，移除了neo cassette。纯化后的转基因被注入C57BL/6雌性卵母细胞的核。产生了两个克隆，1-Sox10和2-Sox10。其中，1号克隆含有2个拷贝的转基因，而2号克隆由于表达水平更高，含有9个拷贝（来源：J:185481）。
