这个靶向载体的设计如下：从5'至3'，包含一个内向面的lox2272:loxP位点，一个反向的Shank3 PDZ域(包括13-16号外显子)，另一个内向面的lox2272:loxP位点，以及一个FRT引导的诺卡因抗性基因座，所有这些都替换了相应的内源性Shank3序列。通过Flp介导的重组，FRT引导的诺卡因基因座被移除。(来源：J:229668)
