The targeting construct replaced 20 kb of endogenous sequence from 5' end of exon 2 to the 5' end of exon 7 with a STOP cassette, IRES, beta-geo and polyadenylation sequence. RNA blotting confirmed the absence of transcript expression in intestinal epithelial cells. Immunohistochemistry confirmed the absence of protein expression in the small intestine and colon.
