在exon 5上游插入了一个loxP位点。随后，插入了一个编码其余外显子、polyadenylation序列、YFP报告基因、FRT位点、诺卡因抗性基因、另一个loxP位点和FRT位点的cDNA。通过Flp介导的重组，去除了片段 cassette，保留了exon 5的loxP标记，并且报告基因完好无损。（来源：J:221922）
