45 nucleotides in exon 2 after the start codon that encode an N-terminal signal sequence were deleted. A floxed stop and neomycin resistance cassette was inserted upstream of the modified exon 2. Cre-mediated recombination is needed to achieve expression of the intracellular isoform. Western blot analysis confirmed the absence of protein expression (intracellular or soluble isoforms) in splenocyte. (J:222609)
