在E18.5期的颅骨细胞中，对II型特异性翻译起始位点在第二外显子进行了替换，用了一个终止密码子(TGA)并插入了一个下游的诺卡因抗性基因片段。RT-PCR结果显示，I型蛋白的表达减少了84%，而II型蛋白的表达则基本保持在40%以上，杂合突变小鼠中这两种表达均显著降低。Runx2的整体表达减少了67%。（来源：J:223410）
