将loxP切割位点插入exon 32的5'端，同时将含FRT引导的诺卡因抗性基因(Neo)和另一个loxP位点的片段插入exon 32的3'端，制备了靶向载体。携带目标等位基因的生殖细胞系小鼠与表达Flp重组酶的鼠杂交，产生了携带删除了exon 32的转基因小鼠。这些带有删除突变的后代再与表达Cre重组酶的鼠交配，以去除exon 32。来源：(J:227953)
