在目标基因的第2外显子(第一个编码外显子)下游，插入了一个含有FRT位点引导的neo cassette和一个loxP位点的靶向载体，同时在第2外显子上游也插入了另一个loxP位点。通过Flp介导的重组，FRT位点引导的neo cassette被移除，使得第2外显子成为可删除的。随后，通过Cre介导的重组，第2外显子被去除。神经组织的western blot检测结果显示，没有蛋白质表达。(来源：J:227400)
