The cDNA encoding human KCNIP3 (DREAM) with two amino-acid substitutions each at EFhands 2, 3 and 4 (EFmDREAM) was inserted downstream of the human CaMKII promoter. The cDNA was followed by an IRES and the LacZ reporter gene. The amino acid substitutions rendered the mutant insensitive to Ca2+, and thus can block the Ca2+dependent derepression function of DREAM and other KChIP proteins endogenously expressed. (J:103733)
