U6-shRNA构建包含了驱动shRNA表达的U6小鼠启动子，该启动子驱动针对Pseudorabies病毒（PRV）中间早期基因3'端的特定序列。目标序列是5'-CTCCCATCGGGTGAGAAAA-3'。Pseudorabies病毒目前感染农场动物，特别是猪和小鼠。U6-shRNA基因插入到一个包含鸡β珠蛋白基因座的5'HS4绝缘子、人类eF1α基因启动子（-1430至+2）、兔（rb）β珠蛋白的第二个内含子以及多个增强子（E1、E2和E3）的空表达载体中，以确保可靠表达。构建了SHO3、SHO5和SH30转基因品系。所有这些品系对PRV病毒感染具有抗性。（来源：J:211915）
