将Fucci2a基因，由CAG启动子控制，通过同源重组插入Rosa26位点。Fucci2a基因包含一个双顺式转录报告细胞周期进展，表达mVenus-hGem(1/110)和mCherry-hCdt1(30/120)。mVenus-hGem是人类Geminin蛋白110个氨基酸N端的融合报告器，mCherry-hCdt1是人类Cdt1蛋白30-120位点的截短版本。这两种蛋白通过Thosea asigna病毒2A(TEV)自切割肽序列融合。构建中的这种融合蛋白在自我切割后应能预测产生等量的细胞周期探针。R26Fucci2aR动物中未观察到Fucci2a表达，因为floxed-Neo-pA的强转录终止序列。经Cre介导重组后，绿色的Venus荧光蛋白和红色的Cherry荧光蛋白在活跃增殖细胞中强烈表达。(来源：J:222845)
