The targeting construct inserted into the locus a CAG promoter, floxed membrane-targeted tandem-dimer Tomato (mT) reporter gene, an improved sensor cGMP indicator (cGi500; containing an FRET-activated, cGMP-sensitive ECFP/EYFP fusion) and an FRT-flanked neomycin resistance cassette. Cre-mediated recombination removed the Tomato reporter gene permanently activating the cGMP sensor allele so that the cGMP sensor is expressed ubiquitously.
