Using BAC recombineering roughly 3 Mbp of the mouse locus including all of the Vk and Jk segments was replaced with 480 kb of human IGK sequence including the proximal Vk and all of the Jk human gene segments. The final selection cassette was removed by flp-mediated recombination. (J:208619)
