使用了BAC(细菌人工染色体)文库(CHORI)来获得一个包含整个小鼠活动调节的细胞骨架关联蛋白(Arc)基因的BAC(编号#RP23-429N15)。随后，插入了一个CreERT2融合基因(连接了人类雌激素受体的配体结合域的Cre重组酶)，后面跟着SV40的polyadenylation序列，这个序列被插入到了基因的起始翻译位点。建立了M和F两个祖先系，并进行了维持(来源：J:215456)。
