插入到定位点的靶向载体设计如下：从5'至3'，包含一个5'同源臂、一个普遍表达的CAG启动子、一段由loxP引导的3xSV40polyA终止子、一个3X-FLAG标签、一个经过哺乳动物优化的Cas9基因（来自链球菌噬菌体CRISPR相关蛋白9，SpCas9）、两个核定位信号、P2A核糖体跳过肽序列、增强型绿色荧光蛋白（EGFP）基因、木瓜病毒后转录调控元件、牛生长激素polyA序列、pPGK-Neo-pA选择性标记片段，以及一个3'同源臂。Cre介导的重组会去除含STOP子的可删除序列。（来源：J:213550）
