设计了一个靶向载体，包含一个单个loxP位点，随后是带有frt内切酶座的诺卡因抗性(neo)基因片段，位于基因的exon 2上游。在exon 2上游，插入了3X Flag序列和另一个loxP位点，位于Nkx2-5的终止密码子之前。通过FLP介导的重组，去除了含frt标记的neo基因片段。（来源：J:101977）
