在exon 5上游插入了floxed neomycin抗性 cassette，然后在exon 5下游插入了另一个loxP位点。通过Cre介导的重组，去除了选择性 cassette和exon 5。这个操作导致催化域的第一个编码exon被消除，并且后续的下游exons产生了框架移位。Northern blot结果显示没有转录本表达。（来源：J:210147）
