A conditional by inversion (COIN) intron consisting of a lox66_SA-Egpf-polyA_lox71 with a dsRed insertion was inserted in the antisense orientation into intron 1 via homologous recombination, splitting intron 1 into 2 new exons (1,036 and 4,595 bp in length). A 2 bp deletion occurs at the site of insertion. The two lox sites are in reverse complementation orientation, thus cre-mediated recombination will irreversibly invert the inserted sequence. In addition, a neomycin resistance cassette was inserted after the the dsRed. Cre-mediated recombination inverts the EGFP cassette to its functional position. (J:200671)
