将cre/EGFP融合基因和Frt末端的Hygro筛选 cassette通过重组合插入到一个Nkx2-2基底交换载体中，使得cre/EGFP与起始密码子ATG成一体，替换掉前导编码exon除最后25个碱基外的所有序列。这个载体与含Nkx2-2(loxed cassette acceptor)基因型的ES细胞共转染。通过正确交换的ES克隆产生嵌合体，然后与B6小鼠杂交来验证遗传传递。接着，这些动物与ACTB-Flpe小鼠杂交以去除Hygro标记。(来源：J:204532)
