BAC克隆RP24-289D15通过ET重组技术改造，插入了包含混合内含子、Cre重组酶、IRES位点和β-半乳糖苷酶（lacZ）序列的元件，以及FRT标记的诺卡因/卡那霉素选择标记，目标插入在预神经肽（Penk）起始编码基因的ATG位置。随后，通过Flp重组酶去除了选择标记。经过纯化并经测序验证的修改后圆形BAC克隆，然后注射到卵母细胞中。目前找不到供体的信息。（来源：J:204429）
