插入载体包含一个带有lacZ截断序列、polyadenylation信号以及GFP标记的人源H2B基因，最后是FRT引导的诺卡因抗性筛选元件，它被插入在exon 1的起始密码子上游6个碱基的位置。在正确靶向的细胞中表达的Flp重组酶会去除这个筛选元件。只有在lacZ截断序列被去除后，标记的H2B基因才会表达。(来源：J:198628)
