设计了一个靶向载体，包含连接有IRES的Cre重组酶，后面跟着氧垂体(Oxt)终止密码子下游的Frt座标记的kanamycin抗性基因。这个载体通过电穿孔法导入了129S6/SvEvTac衍生的W4胚胎干细胞(ES细胞)。经过定向的ES细胞被注入囊胚，然后杂合子小鼠与表达flp的个体杂交，以去除kanamycin选择性基因组。(来源：J:192007)
